Firefly Luciferase (FLuc) mRNA is a highly sensitive reporter commonly used to evaluate mRNA delivery, transfection efficiency, and in vivo protein expression. The data demonstrate representative results obtained using RiboPro’s FLuc mRNA in both animal and cell-based transfection studies.
Bioluminescence imaging performed 6 hours after intravenous administration of FLuc mRNA encapsulated in lipid nanoparticles (LNPs) showed robust luciferase expression in C57BL/6 mice. Following injection of 0.5 mg/kg FLuc mRNA, strong bioluminescent signals were detected, demonstrating efficient in vivo delivery and protein expression.
In vitro transfection studies in bone marrow-derived dendritic cells (BMDCs) further confirmed the performance of FLuc mRNA. Luciferase activity was measured after 24 and 48 hours following delivery using PLGA/PBAE nanoparticles, PBAE nanoparticles, or Lipofectamine 2000. All delivery systems mediated detectable expression, with Lipofectamine 2000 producing the highest luciferase activity, while PLGA/PBAE formulations showed substantial reporter expression compared to untreated controls.
